چکیده

NEUROTROPHIN EXPRESSION IN SYMPATHETIC NEURONS: INFLUENCES OF EXOGENOUS NGF AND AFFERENT INPUT by Elizabeth Ellen Jones The regulatory mechanisms that promote the survival of adult neurons remain unclear. Targetderived proteins called neurotrophins are known to play a role in the survival of sympathetic neurons, but their exact function is unknown. In the present study, expression of the neurotrophins nerve growth factor (NGF) and neurotrophin-3 (NT-3) in adult sympathetic ganglia and their targets was investigated. Using Western blot analysis, multiple precursor forms of both NGF and NT-3 were observed in neurons and blood vessel targets. In addition, using real time RT-PCR, transcripts for both NGF and NT-3 were detected in the ganglia, demonstrating the potential for neurotrophin synthesis in peripheral ganglia. The expression patterns of both NGF and NT-3 were found to be altered by both exogenous NGF and removal of pre-ganglionic input, suggesting the regulatory influences of both NGF and afferent input on neurotrophin expression and ultimately neuronal survival. Overall, this study advanced the understanding of neurotrophin regulation of neuronal survival NEUROTROPHIN EXPRESSION IN SYMPATHETIC NEURONS: INFLUENCES OF EXOGENOUS NGF AND AFFERENT INPUT

Specific high-affinity receptors for neurotrophin-3 on sympathetic neurons.

When used at concentrations allowing interactions only with its high-affinity receptors, neurotrophin-3 (NT-3) promotes the survival of sensory neurons isolated from embryonic day 8 (E8) chicks, but not the survival of E11 sympathetic neurons. These sympathetic neurons (which can be rescued by the addition of NGF) display high-affinity receptors for NT-3 (Kd of 1.6 x 10(-11) M) that cannot be d...

Neurotrophins alter the numbers of neurotransmitter-ir mature vagal/glossopharyngeal visceral afferent neurons in vitro.

Mature nodose and petrosal ganglia neurons (placodally derived afferent neurons of the vagal and glossopharyngeal nerves) contain TrkA and TrkC, and transport specific neurotrophins [nerve growth factor (NGF), neurotrophin-3 (NT-3), neurotrophin-4 (NT-4)]. This study evaluated neurotrophin influences on the presence of neuropeptides and/or neurotransmitter enzymes in these visceral sensory neur...

Loss of neurofibromin results in neurotrophin-independent survival of embryonic sensory and sympathetic neurons

Mutations at the neurofibromatosis 1 (NF1) locus in humans and mice result in abnormal growth of neural crest-derived cells, including melanocytes and Schwann cells. We have exploited a targeted disruption of the NF1 gene in mice to examine the role of neurofibromin in the acquisition of neurotrophin dependence in embryonic neurons. We show that both neural crest- and placode-derived sensory ne...

Absence of the p75 neurotrophin receptor alters the pattern of sympathosensory sprouting in the trigeminal ganglia of mice overexpressing nerve growth factor.

Sympathetic axons invade the trigeminal ganglia of mice overexpressing nerve growth factor (NGF) (NGF/p75(+/+) mice) and surround sensory neurons having intense NGF immunolabeling; the growth of these axons appears to be directional and specific (). In this investigation, we provide new insight into the neurochemical features and receptor requirements of this sympathosensory sprouting. Using do...

Phenotypic modification of primary sensory neurons: the role of nerve growth factor in the production of persistent pain

Inflammation results in an early and maintained elevation in nerve growth factor (NGF) levels in inflamed tissues. Neutralizing the action of the increased NGF with specific anti-NGF antibodies substantially diminishes inflammatory hypersensitivity, indicating that this neurotrophin is a key mediator in the production of inflammatory pain. The hyperalgesic actions of NGF may in part be the cons...
